Owing to deep research experience in HBV envelop, Beacle launches quantitative ELISA kits targeted to varous HBsAg domains. Three types of HBsAg is known; named S-, M- and L-antigen. L-antigen is consisted of S, Pre-S2 and Pre-S1 domains, M-antigen of S and Pre-S2 domains, and S-antigen of S domain alone. These domains play important role in HBV infection. Pre-S1 domain is known to be the key element in recognizing hepatic cells, Pre-S2 and S is important in cellular invasion fo HBV. The serum level of S-antigen (or S-domain) is well studies as a tool to daignose the HBV infection, but almost no study was conducted to examine the serum level of Pre-S1 and Pre-S2. Beacle, by marketing ELISA kit to detect these domains, wishes to contribute studies of HBV infection.

For HB Pre-S1 Antigen Quantitative ELISA Kit
Q: How long the kit can preserve?
All the component of the kit is stable for at least 6 months at 4℃.
Q: How long does it take for an assay
The approximate time required for an assay is shown below.
1) Antigen ELISA kits: Reaction with capture IgG and detection IgG: 60 min→5) Chromogenic reacton: 30 min
2) Antibody ELISA kits: Reaction with capture antigen blocking, and detection IgG: 2 hrs→5) Chromogenic reacton: 30 min
Q: What is the definition of PreS-1 activity？
There is no common and standard definition of Pre-S1 antigen activity. In the kit the activity is expressed by our unit system, where 1 nUnit equal to the Pre-S1 activity expressed by 1ng of our standard antigen. For S antibody, our kit uses mouse IgG for standard. The binding analysis of the two standard revealed that 10mIU/mL of WHO standard serum is roughly equivalent to about 1nUnit/mL. Please remember that the estimation is not accurate because the binding property of the standard antibody to S antigen is different from that of WHO standard serum in both Bmax and Kd.
Q: What is the standrd antigen or standard antibody？
The standard antigen are the recombinant HBsAgs produced from yeast. These antigens gives reproducible antigen activity by antigen ELISA. For antibody ELISA mouse monoclonal antibody is provided for standard antibody in kit for mouse
Q: How can I calculate the sample concentration？
The sample concentration is calculated based on four parameter fit logistic model. Usually Plate leader is equipped by such program, and we recommend to use the program. If you do not have such program, you can use a sample Excel program.
